Solanum nigrum L. (Köpek Üzümü)’un Kuraklık ve Tuz Stresine Fizyolojik Tepkilerinin Karşılaştırılması

Abstract

Kuraklık ve tuzluluk, küresel iklim değişikliğinin bir sonucu olarak ortaya çıkan başlıca çevre sorunları arasında yer almaktadır. Bu çalışmada, istilacı bir yabancı ot olan köpek üzümünün (Solanum nigrum L.) değişen, gelişen kuraklık ve tuzluluk koşullarına tepkisinin antioksidan kapasite temelinde tepkisinin belirlenmesi amaçlanmıştır. Bu amaçla, Solanum nigrum tohumları petrilerde çimlendirilip perlit içeren viyollere aktarılmıştır. Fideler 42 gün boyunca Hoagland Besin Solüsyonu (%100) ile in vitro şartların sağlandığı bitki büyütme kabininde yetiştirilmiştir. Daha sonra fideler kontrol (C), kuraklık stresi (D) ve tuz stresi (100 mM NaCl) olarak üç gruba ayrılmıştır. Bir haftalık uygulamanın ardından 7. günde fidelerden örnekleme yapılmıştır. Örneklerde bağıl su içeriği (BSİ), kuru ağırlık (KA), spesifik yaprak alanı (SYA) ve hidrojen peroksit (H2O2) miktarı ölçülmüştür. Kuraklık ve tuzluluk stresi uygulamaları ile BSİ özellikle kuraklık stresiyle kontrole kıyasla %27 azalmıştır. Kuru ağırlık ise kuraklık stresiyle %54, tuz stresiyle %41 azalmıştır. Ayrıca H2O2miktarı da kuraklıkla %72, tuzlulukla %48 azalmıştır. Elde edilen bulgular, köpek üzümünün tuzluluğa kıyasla kuraklıktan daha çok etkilendiğini işaret etmektedir.

Comparison of Physiological Responses of Solanum nigrum L. (Blackberry Nightshade) to Drought and Salt Stress

Abstract

Drought and salinity are among the main environmental problems because of global climate change. Our aim of this study to determine the physiological and biochemical responses of black nightshade (Solanum nigrum L.) under drought and salt stress conditions based on antioxidant capacity. For this purpose, the seeds of Solanum nigrum were germinated in petri dishes and then transferred to viols containing perlite. The seedlings were grown in a plant growth cabinet with Hoagland Nutrient Solution (100%) for 42 days in vitro conditions. The seedlings were then divided into three groups: Control group (C), drought stress group (D) and salt stress group (100 mM NaCl). Relative water content (RWC), dry weight (DW), specific leaf area (SLA) and hydrogen peroxide (H2O2) amount were measured from the seedlings on the 7th day after one week of application. With drought and salinity stress applications, RWC decreased by 27% with drought stress compared to control. The decrease in dry weight was determined as 54% with drought stress and 41% with salt stress. In addition, the amount of H2O2decreased by 72% at drought and 48% at salinity condition. Results of the study indicated that blackberry nightshade is more affected by drought compared to salinity.
